I've done it warm a couple of times, just using cells that were
subcultured ~6 hours before, then pelleted and resuspended in water a
few times before adding plasmid DNA and zapping with the MicroPulser.
I have also done it cold, but those times I was using
"electrocompetent" cells which were rinsed with much finer fluids than
plain water... so they had higher efficiencies, but I have never had
arcing in any case (though I can't quite remember what happened when I
tried using a grill lighter to electroporate, heh heh).
